表皮葡萄球菌调节信号多肽及编码的DNA 【技术领域】
本发明属生物技术领域,具体涉及作为表皮葡萄球菌信号传递系统关键新蛋白:组氨酸蛋白激酶和反应调节蛋白,以及利用该类蛋白寻找抑菌药物的方法。更具体地,本发明涉及编码该类多肽的DNA;制备该类多肽的方法,包括基因克隆、基因表达、蛋白纯化和功能测定;制备该类多肽的抗体;以及利用该类多肽筛选抑菌活性化合物。
背景技术
葡萄球菌与医院内化脓性感染相关,尤以金黄色葡萄球菌和表皮葡萄球菌最为密切,表皮葡萄球菌所致感染与一些高分子医疗器材在体内的留置有关,如静脉导管、起搏器、脑脊液引流装置、人工器官等。随着上述医疗材料的使用日益频繁,此类院内感染问题日益严峻,且易发展为慢性感染,对抗生素的抗性较高。
已知在各种原核生物中普遍存在双组分调控系统,为原核细胞所特有的信号系统,参与调控生物生长、毒力等多种生物学功能。通常由一对相应的蛋白组成,称为组氨酸蛋白激酶和反应调节蛋白。组氨酸蛋白激酶为跨膜蛋白,当外界信号作用于组氨酸蛋白激酶的膜外区域,产生信号,使该蛋白发生自身磷酸化。随后,磷酸化的组氨酸蛋白激酶作用于相应的反应调节蛋白发挥调控作用。不同的双组分蛋白,调控细菌的不同生物学活性。然而,由于研究手段的局限性,对表皮葡萄球菌双组分调控系统及对生物学功能地调控缺乏系统地了解和实际应用。
【发明内容】
本发明的目的是提供一组表皮葡萄球菌信号传递系统的新蛋白,包括组氨酸蛋白激酶和反应调节蛋白。本发明的另一目的是提供制备该类多肽的方法和编码该类多肽的DNA,及其制备该类多肽的抗体,本发明的进一步目的是利用该类多肽筛选药物化合物。
本发明通过对表葡菌ATCC12228株的全基因组测序和序列分析,采用序列同源性比较和功能域分析等生物信息学方法确定了16对表皮葡萄球菌的双组分调控信号蛋白(包括组氨酸蛋白激酶和反应调节蛋白),并制备了相应的蛋白,经对上述蛋白功能域空间结构的模建和实验证实该类蛋白存在保守性结构和活性位点,可作为筛选药物的“靶点”。
本发明表皮葡萄球菌组氨酸蛋白激酶和反应调节蛋白中包括16对双组分调控蛋白,其中16个组氨酸蛋白激酶均具有结合ATP的激酶活性功能域(HATPase_c功能域),分别具有SEQ ID No.1-16的序列结构。其中16个反应调节蛋白均存在接受磷酸基团的功能域(REC功能域),分别具有SEQ ID No.17-32的序列结构。
本发明采用PMODELIHG软件对上述相对保守的功能域片段进行同源模建,结果显示:组氨酸蛋白激酶HATPase_c功能域的结构特征是在AMP-PNP的结合部位形成袋状结构,且在其周围有保守的基序围绕,所述AMP-PNP是一种ATP类似物;反应调节蛋白的REC功能域模拟结构由5个α螺旋和5个β折叠组成,其中存在三个天冬氨酸残基构成接受磷酸基团的活性位点,且接受磷酸基团的过程需要二价阳离子(Mg2+或Mn2+)的辅助,所述天冬氨酸残基是D7、D8、D51。
本发明所述的表皮葡萄球菌组氨酸蛋白激酶具有SEQ ID No.1-16的氨基酸序列结构,由SEQ ID No.33-48的DNA核苷酸序列编码;所述的反应调节蛋白具有SEQ ID No.17-32的氨基酸序列结构,由SEQ ID No.49-64的DNA核苷酸序列编码。
本发明涉及由SEQ ID No.1-32构成的蛋白的氨基酸序列的同源序列及片断序列,和SEQ ID No.33-64编码相关同源序列和片断有关多肽的DNA核苷酸序列。所述同源DNA序列是指编码SEQ ID No.33-64所示核苷酸序列或片断,或DNA片断长度在60个连续核苷酸区域内,一般至少70%同源于SEQ ID 1-32,可与之选择性杂交的片断核苷酸序列。所述SEQ ID 1-32的多肽同源序列至少在30个的连续氨基酸序列区域,至少60%以上的同源。
本发明的多肽包括组氨酸蛋白激酶和反应调节蛋白氨基酸序列的功能域部分序列,如具有生物学活性的氨基酸基本序列的多肽片断;或其中部分氨基酸被性质相似的氨基酸取代的多肽;或其氨基酸的一部分中加入或插入其他氨基酸的多肽,以及DNA的核苷酸序列改变,但不影响氨基酸序列的多肽,如同种氨基酸使用不同密码子。
本发明提供了制备表皮葡萄球菌组氨酸蛋白激酶和反应调节蛋白(SEQ IDNo.1-32)方法,包括制备仅含生物学活性功能区的编码序列的DNA片断,克隆至适合的表达载体,转染宿主菌,在适于的条件下培养该宿主细胞,以及从培养物中回收和纯化所需蛋白。
上述技术方案中所有基本分子生物学操作均参照《分子克隆实验指南》。
本发明将编码相应蛋白的DNA序列与表达载体重组,形成重组表达质粒。本发明不限于特定的表达质粒。在一优选实施方案中,本发明使用原核表达载体,例如pet28a等。
上述重组表达载体可按常规方法导入适宜宿主细菌。本发明不局限于任何特定的宿主细胞,只要它能够表达所述重组表达载体。在一优选实施方案中,本发明使用大肠杆菌JM109等。
本发明的多肽包括磷酸化反应产物。
本发明还提供抗本发明多肽的多克隆抗体或单克隆抗体,使用本发明的多肽或其片断作为免疫原,按常规方法制备特异性抗体。可用于细菌细胞内的多肽表达量,也可用于研究所述多肽与细菌致病的关系,或用于阻断细菌细胞之间的信号转递。
可以将本发明DNA的反义核酸的DNA插入合适的载体中,产生反义RNA;也可用合成方法产生反义RNA。反义RNA可用于控制细菌细胞内多肽水平。
本发明多肽功能域结构可用于筛选活性化合物,能阻断组氨酸蛋白激酶或反应调节蛋白质间的磷酸基团转移和蛋白质的磷酸化,抑制细菌细胞内或细胞之间的信号传递,从而抑制葡萄球菌的生长。
本发明在表皮葡萄球菌ATCC12228株中确定了16对完整的双组分调控系统的基因核苷酸序列及其功能。经实验证实了组氨酸蛋白激酶和反应调节蛋白的磷酸化或两者分子之间的磷酸基团转移可调控葡萄球菌的生物学活性,抑制组氨酸蛋白激酶和反应调节蛋白的磷酸化或磷酸基团的转移可抑制细菌的生长。对其保守性功能域的空间结构模建确定了存在保守结构和活性位点可作为筛选活性化合物的药物“靶点”。

【具体实施方式】
实施例1 确定16对表皮葡萄球菌双组分调控系统
运用序列同源性比较,功能域分析等生物信息学方法,在表皮葡萄球菌ATCC12228株全基因组(GeneBank收录号AE015929)中确定了16对双组分调控基因。表1列出了上述双组分调控基因的基本情况,包括基因长度等,以及氨基酸序列同源性比较结果(blastp)。

通过与SMART氨基酸序列及功能域数据库进行同源性比较,对16对双组分调控基因中存在的保守性功能域进行分析,结果表明16个组氨酸蛋白激酶中均存在结合ATP的激酶活性功能域(HATPase_c功能域),16个反应调节蛋白(SEQID No.17-32)均存在接受磷酸基团的功能域(REC功能域),表2是表皮葡萄球菌双组分调控基因功能域分析结果。

实施例2 对HATPase_c功能域和REC功能域进行空间结构模建
根据上述确定的HATPase_c功能域和REC功能域在双组分调控系统中的相对保守性,运用PMODELING软件对两个功能域进行空间结构的同源模建。结果显示HATPase_c功能域模拟结构,与已报道的大肠杆菌组氨酸蛋白激酶基因EnvZ的HATPase_c功能域的和磁共振(NMR)结构非常相似(Ncbi mmdbId:11091),即HATPase_c功能域主要结构由4个β折叠和3个α螺旋组成,尤其是在AMP-PNP(一种ATP类似物)的结合部位形成一个袋状结构,且其周围有一些保守的基序(G1 box、G2 box、F box)围绕,见图1a;而REC功能域模拟结构与大肠杆菌的反应调节蛋白基因CheY的REC功能域的和磁共振(NMR)结构也非常相似(NcbimmdbId:3000),由5个α螺旋和5个β折叠组成,其中存在三个天冬氨酸残基(D7、D8、D51)构成接受磷酸基团的活性位点,且该过程需要二价阳离子(Mg2+或Mn2+)的辅助,见图1b。因此,这两个结构均存在药物设计、筛选的“靶标”,以它们为模板,筛选各类小分子化合物库,能找到一些在空间结构上封闭该活性结构或活性位点的小分子化合物,从而阻断这些双组分调控系统,其作用原理是阻断双组分调控系统信号传导途径的一个或多个环节,包括组氨酸蛋白激酶的自身磷酸化、将磷酸基团从蛋白激酶转移到调节蛋白的转磷酸基过程以及反应调节蛋白的磷酸化等。
实施例3 16对双组分调控基因相关功能的分析
将16对双组分调控基因与已报道的其他革兰阳性菌中有明确功能的双组分基因进行了序列同源性比较,结果发现有8对双组分调控基因与已报道的其他革兰阳性菌中双组分基因有较高的氨基酸序列同源性(同源性大于30%),表明所述双组分调控系统在同为革兰阳性菌的表皮葡萄球菌中也可能发挥着重要的作用,表3是表皮葡萄球菌双组分蛋白调控的生物学功能同源性比对结果。

(2)重组质粒1175-pet28a、1176-pet28a、1175d-pet28a的表达及目的蛋白的纯化:
将上述重组质粒1175-pet28a、1176-pet28a、1175d-pet28a按常规方法在IPTG诱导下表达,用SDS-PAGE电泳对目的蛋白进行鉴定,结果显示三种重组质粒分别表达70kd、30kd及30kd左右的蛋白,用申能博彩公司提供的3s NTAHis-Tag Resin层析柱进行蛋白提纯。
